Genetic mapping PROLIFERA has been mapped to genetic marker GT148, at a position of 11.6 cM from the top arm of chromosome IV.

Research published by Springer PS, et al. (1995, Science 268:877-880) describes the PROLIFERA (PRL) gene as required for megagametophyte and embryo development. PRL was identified by gene trap transposon mutagenesis and is related to the MCM2-3-5 family of yeast genes required for the initiation of DNA replication. Heterozygotes for disruption of PRL are semi-sterile; siliques contain mutant embryo sacs arrested at various stages: unfertilized ovules and aborted embryos. This suggested reduced viability of the megagametophyte and was confirmed by reciprocal crosses to wild-type plants. A 2.7 kb transcript was detected in RNA from roots, buds, and flowers, while GUS staining indicated that PRL is expressed in dividing cells throughout the plant.
